今日からは気分も新たに新テーマです

バリデーションにおけるテスト結果の記録方法について、具体的な方策でわかりやすく説明している英語の記事を見つけたのでシリーズでご紹介しようと思います。

製薬会社さんにとっては、CSVでも通常業務の記録方法と同等にテスト結果を記録すると言えばどうすればよいか想像つくかなと思いますが、供給者(システム開発会社)は「製薬業界での標準的な記録方法」が良く分からないため、対応に困ってしまうことがあるのではないでしょうか。薬会社さんにとっても、どのように供給者に依頼すればよいかという点が悩みどころかと思います。

最近では、システムテストや受入試験など、開発フェーズで供給者が実施するテスト結果をOQやPQに引用したりすることも認められてきていますので、供給者によるテスト記録の作成が重要になってきています。

そこで今回のシリーズを参考に、バリデーションにおけるテスト結果の記録方法を再確認していってもらえればと思います。

まず初めに、筆者のご紹介です。
今回の記事の筆者は、Don Hurdさんです。

Selection of a container and packaging should be based on the storage and transportation requirements of the medicinal products; the space required for the amount of medicines; the anticipated external temperature extremes; the estimated maximum time for transportation including transit storage at customs; the qualification status of the packaging and the validation status of the shipping containers.
容器及び包装の選択は、次に挙げる医薬品の保管及び輸送要件に基づいていなければならない。

9.3.3
Containers should bear labels providing sufficient information on handling and storage requirements and precautions to ensure that the products are properly handled and secured at all times. The containers should enable identification of the contents of the containers and the source.
容器には、製品が常に適切に取り扱われ、保護されることを保証するために取り扱い及び保管要件に関する十分な情報及び使用上の注意を表示しなければならない。 



9.4 PRODUCTS REQUIRING CONTROLLED CONDITIONS

9.4.1
In relation to deliveries containing medicinal products requiring special conditions such as narcotics or psychotropic substances, the wholesale distributor should maintain a safe and secure supply chain for these products in accordance with requirements laid down in national legislation. There should be additional control systems in place for delivery of these products. There should be a protocol to address
the occurrence of any theft.
麻薬や向精神薬のような特殊な条件を要する医薬品の配送では、卸売業者は、各国の法令で定められている要件に従って、該当製品の流通経路を安全確実に維持しなければならない。該当製品の配送においては、付加的な管理システムを構築しなければならない。盗難発生時の対応に関する手順を整備しなければならない。

9.4.2
Medicinal products comprising highly active and radioactive materials should be transported in safe, dedicated and secure containers and vehicles. The relevant safety measures should be in accordance with international agreements and national legislation.
高活性及び放射性物質から成る医薬品は、専用の保護された容器及び車両で安全に輸送しなければならない。関連する安全対策は、国際協定及び各国の法令に従って実施しなければならない。。

9.4.3
For temperature-sensitive products, qualified equipment (e.g. thermal packaging, temperature-controlled containers or temperature controlled vehicles) should be used to ensure correct transport conditions are maintained between the manufacturer, wholesale distributor and customer.

If temperature-controlled vehicles are used, the temperature monitoring equipment used during transport should be maintained and calibrated at regular intervals. Temperature mapping under representative conditions should be carried out and should take into account seasonal variations, if applicable.

If requested, customers should be provided with information to demonstrate thatproducts have complied with the temperature storage conditions.
要求に応じ、製品が保管温度条件内にあったことを示す情報を顧客に提供しなければならない。

9.4.6
If cool packs are used in insulated boxes, they need to be located such that the product does not come in direct contact with the cool pack. Staff must be trained on the procedures for assembly of the insulated boxes (seasonal configurations) and on the reuse of cool packs.
断熱容器に保冷剤を使用する場合は、保冷剤が製品に直接接触しないように配置しなければならない。従業員は(季節に応じた)断熱容器の組み合わせ組み立て及び保冷剤の再利用について訓練を受けなければならない。

There should be a system in place to control the reuse of cool packs to ensure that incompletely cooled packs are not used in error. There should be adequate physical segregation between frozen and chilled ice packs.

前回から引き続いて「9章 輸送」を見てきましたが、JGSP(※3)でも同様の目的で「2.配送業務」があります。

JGSPと比べると、PIC/S GDPでは医薬品が適切な状態であったことを実証することが強く求められているように思います。つまり、手順を確立し、その手順を遵守して記録を残すことが全般に的に言われています。特に温度管理や保管容器、保冷剤などについては、バリデーションをして保証することになります。

と言われても、何から手を付けてよいかわからない、、、という方は、まずは温度計の管理から始めてみてはいかがでしょう。

PIC/S GDPでは常に正しい温度を記録できるように定期的に校正を実施するよう言われています。リスクベースドアプローチに基づいて、どこで厳密な温度管理が必要となるか優先順位をつけ、そこで使用されている温度計の管理手順書を作成し、定期的に校正する仕組みを作っていく、というような流れになるのではないかなと思います。

こんにちは
気付けば西日が差し込む時間となってしまいました。

昨日、愛読のYahoo!ニュースで恐ろしいニュースが掲載されました。

いろいろな場所に設置されている監視カメラ、設置した人が買った時のままパスワードを変更せずにいると、世界中の人に見られてしまうようです
そして、ロシアのあるサイトでは、ご親切にもそのような監視カメラの情報を集め、公開してくれています

怖いもの見たさで少し覗きに行ったら、日本の監視カメラの映像が非常に多く公開されていました。なんと、アメリカに次いで2番目の数です。

皆さん気を付けてくださいね、と言いたいところですが、私自身も普段はあまり監視カメラの位置を意識して生活していないので、気付かないうちに全世界に公開されているってこともありそうです。

とりあえず、エレベータでは一人しか乗っていなくても外向けを装っていようと思います。

9.1 PRINCIPLE
9.1 原則

9.1.1
It is the responsibility of the supplying wholesale distributor to protect medicinal products against breakage, adulteration, theft and to ensure that temperature conditions are maintained within acceptable limits during transport.

供給元となる卸売業者は、医薬品を破損、不純物の混入、盗難から保護し、輸送期間において温度を許容範囲内に維持することに対する責任を負う。

9.1.2
Regardless of the mode of transport, it should be possible to demonstrate that the medicines have not been exposed to conditions that may compromise their quality and integrity. A risk-based approach should be utilised when planning transportation.
輸送方式にかかわらず、品質と完全性を低下させる恐れのある状態に医薬品がさらされていなかったことを実証できなければならない。輸送の計画を立てる際は、リスクベースドアプローチを活用しなければならない。 


9.2 TRANSPORTATION
9.2 輸送

9.2.1
The required storage conditions for medicinal products should be maintained during transportation within the defined limits as described on the outer packaging and/or relevant packaging information.
医薬品の保管条件は、外包装に表示されている既定の条件及び/又は関連の包装情報に従って
輸送中も維持されなければならない。

9.2.2
If a deviation such as temperature excursion or product damage has occurred during transportation, this should be reported to the distributor and recipient of the affected medicinal products. A procedure should also be in place for investigating and handling temperature excursions.
輸送中に発生した温度逸脱や製品破損等の逸脱については、当該医薬品に関わる卸売業者及び受領者に報告しなければならない。温度逸脱の調査及び取り扱いに関する手順書を整備しなければならない。

9.2.3
It is the responsibility of the wholesale distributor to ensure that vehicles and equipment used to distribute, store or handle medicinal products are suitable for their use and appropriately equipped to prevent exposure of the products to conditions that could affect their quality and packaging integrity.
卸売業者は、医薬品の配送、保管及び取り扱いにて使用する車両及び設備が、用途に適しており、品質及び包装の完全性に影響を及ぼす状態に製品をさらすことがないよう適切に装備されていることに責任を持つこと。

9.2.4
There should be written procedures in place for the operation and maintenance of all vehicles and equipment involved in the distribution process, including cleaning and safety precautions.
清掃や安全対策を含む配送過程に関わる全ての車両及び設備について、操作及び維持するための手順書を整備しなければならない。

9.2.5
Risk assessment of delivery routes should be used to determine where temperature controls are required. Equipment used for temperature monitoring during transport within vehicles and/or containers, should be maintained and calibrated at regular intervals.
温度管理が必要となる場所を決定する際には、配送ルートのリスク評価を行わなければならない。輸送中における車両内及び/又は容器内の温度監視にて使用する装置は、定期的な間隔で校正し、維持しなければならない。

9.2.6
Dedicated vehicles and equipment should be used, where possible, when handling medicinal products. Where non-dedicated vehicles and equipment are used procedures should be in place to ensure that the quality and integrity of the medicinal product will not be compromised.
医薬品を取り扱う際は、可能であれば、専用の車両及び設備を使用すること。専用ではない車両や設備を利用する場合は、医薬品の品質及び完全性が低下しないことを保証するために手順書を整備しなければならない。

9.2.7
Deliveries should be made to the address stated on the delivery note and into the care or the premises of the consignee. Medicinal products should not be left on alternative premises.
配送記録に記載されている住所へ、荷受人宛又は荷受人の建物まで配送しなければならない。医薬品を別の建物に放置してはならない。

9.2.8
For emergency deliveries outside normal business hours, persons should be designated and written procedures should be available.
営業時間外の緊急配送に備え、担当者を指名し、利用可能な手順書を整備しておかなければならない。

9.2.9
Where transportation is performed by a third party, the contract in place should encompass the requirements of Chapter 7. Transportation providers should be made aware by the wholesale distributor of the relevant transport conditions applicable to the consignment. Where the transportation route includes unloading and reloading or transit storage at a transportation hub, particular attention should be paid to temperature monitoring, cleanliness and the security of any intermediate storage facilities.
第三者が輸送を行う場合は、7章の要件を網羅して契約を交わさなければならない。輸送業者は受託品に関する輸送条件について、卸売業者より通知されなければならない。荷下ろしや再積み込み又は輸送中継地での一時保管では、中間保管施設の温度監視、清浄度及び安全対策に関して、特に注意を払わなければならない。

Provision should be made to minimise the duration of temporary storage while awaiting the next stage of the transportation route.
次段階の配送ルートへ引き渡されるまでの一時的な保管を最短とするために、対策を講じなければならない。

国民的アイドルの次は、一般市民のお話に戻り、、、

今朝は溶けきらずに残っていた雪のせいで、通勤中に何度か滑ってしまいました

Self-inspections should be conducted in order to monitor implementation and compliance with GDP principles and to propose necessary corrective measures.

8.2 SELF-INSPECTIONS

8.2.1 A self-inspection programme should be implemented covering all aspects of GDP and compliance with the regulations, guidelines and procedures within a defined time frame. Self-inspections may be divided into several individual self-inspections of limited scope.
GDPのあらゆる側面について、規制、ガイドライン、手順書を遵守して、定められた時間枠内で自己点検を実施しなければならない。自己点検を複数に分けて、限定した範囲内で個別に実施してもよい。


8.2.2 Self-inspections should be conducted in an impartial and detailed way by designated competent company personnel. Audits by independent external experts may also be useful but may not be used as a substitute for self-inspection.
自己点検は、社内で指定され、能力のある者によって、公平かつ詳細な方法で実行されなければならない。外部専門家による独立した監査は有用であるが、自己点検の代わりとすることはできない。


8.2.3 All self-inspections should be recorded. Reports should contain all the observations made during the inspection. A copy of the report should be provided to the management and other relevant persons. In the event that irregularities and/or deficiencies are observed, their cause should be determined and the corrective and preventive actions (CAPA) should be documented and followed up.
すべての自己点検を記録しなければならない。報告書には自己点検中になされたすべての指摘事項を含まなければならない。不正及び/又は不備が観察された場合には、原因を特定し、是正処置、予防処置(CAPA)を文書化し、追跡調査しなければならない。

製薬会社さんなど、GxPの中でお仕事されている方にとっては、いつもの「自己点検」です。そのため、現在お持ちの自己点検の手順書をほぼそのまま採用することで、この章の対応は可能かなと思います。

一方、GxP未経験の方は、「自己点検」という言葉から本来の目的を誤解してしまうかもしれないため、気を付けてください。
「自己点検」というと、担当者が自身で実施した作業を点検・確認するイメージを持ってしまうかもしれませんが、ここでの「自己点検」とは、どちらかというと「内部監査」に近い内容となります。

具体的には、対象業務を理解している責任者や自己点検の担当者が、規制要件や社内規定通りに業務が行われていることを、事前に作成したチェックリストなどを用いて確認することとなります。

7.1 PRINCIPLE

Any activity covered by the GDP Guide that is outsourced should be correctly defined, agreed and controlled in order to avoid misunderstandings which could affect the integrity of the product. There must be a written Contract between the Contract Giver and the Contract Acceptor which clearly establishes the duties of each party.
GDPガイドの対象となる外部委託業務は、製品の完全性に影響を及ぼす誤解を回避するため、正しく定義し、合意し、管理しなければならない。契約委託者と契約受託者の間で、それぞれの責務を明確に定めた契約書を交わす必要がある。



7.2 CONTRACT GIVER

7.2.1
The Contract Giver is responsible for the activities contracted out.
契約委託者は、外注した活動について責任を負う。


7.2.2
The Contract Giver is responsible for assessing the competence of the Contract Acceptor to successfully carry out the work required and for ensuring by means of the contract and through audits that the principles and guidelines of GDP are followed. An audit of the Contract Acceptor should be performed before commencement of, andwhenever there has been a change to, the outsourced activities. The requirement for audit and frequency should be defined based on risk depending on the nature of the outsourced activities. Audits should be permitted at any time.
契約委託者は、受託者が求められる作業を適切に実施する能力の評価を行う責任があり、また、監査を通じてGDPの原則及びガイドラインが遵守されることに対して責任を負う。契約受託者に対する監査は、委託業務の開始または変更前に実施しなければならない。監査の必要性と頻度は、委託業務の性質によってリスクを基に定めなければならない。


7.2.3
The Contract Giver should provide the Contract Acceptor with all the information necessary to carry out the contracted operations in accordance with the specific product requirements and any other relevant requirements.
契約委託者は、特殊な製品の要件やその他の関連要件に従って、契約業務を実施する上で必要となるあらゆる情報を契約受託者に提供しなければならない。





7.3 CONTRACT ACCEPTOR

7.3.1
The Contract Acceptor is responsible for the activities covered by GDP and delegated by the Contract Giver.
契約受託者は、GDP対象である契約委託者によって委託された業務について責任を負う。


7.3.2
The Contract Acceptor should have adequate premises and equipment, procedures, knowledge and experience, and competent personnel to carry out the work ordered by the Contract Giver.
契約受託者は、契約委託者から発注された業務を実施するための適切な構造設備、知識及び経験、ならびに有能な人員を有していなければならない。


7.3.3
The Contract Acceptor should not pass to a third party any of the work entrusted to him under the contract without the Contract Giver’s prior evaluation and approval of the arrangements and an audit of the third party by the Contract Giver or the Contract Acceptor. Arrangements made between the Contract Acceptor and any third party should ensure that the wholesale distribution information is made available in the same way as between the original Contract Giver and Contract Acceptor.

PIC/S GDPでは医薬品の品質保証や偽造医薬品の混入回避の観点などから、外部委託についてもしっかり管理するように要求されています。
